Reduced Incidence and Severity of Antigen-induced Autoimmune Diseases in Mice Lacking Interferon
نویسندگان
چکیده
Interferon regulatory factor-1 (IRF-1) is a transcription factor that regulates interferon-induced genes and type I interferons. Recently, studies of IRF-1-deficient mice have revealed that IRF-1 regulates the induction of molecules that play important roles in inflammation, such as inducible nitric oxide synthase (iNOS) and interleukin-1 b -converting enzyme (ICE). To study the role of IRF-1 in autoimmunity, we investigated type II collagen-induced arthritis (CIA), and experimental allergic encephalomyelitis (EAE), in mice lacking IRF-1. The incidence and severity of CIA were significantly decreased in IRF-1 2 / 2 mice compared with IRF-1 1 / 2 mice, as was the production of interferon (IFN)g in lymph node cells. Both IRF-1 1 / 2 and IRF-1 2 / 2

Reduced Incidence and Severity of Antigen-induced Autoimmune Diseases in Mice Lacking Interferon Regulatory Factor-1
Interferon regulatory factor-1 (IRF-1) is a transcription factor that regulates interferon-induced genes and type I interferons. Recently, studies of IRF-l-deficient mice have revealed that IRF-I regulates the induction of molecules that play important roles in inflammation, such as inducible nitric oxide synthase (iNOS) and interleukin-l beta-converting enzyme (ICE). To study the role of IRF-1...
متن کاملTolerizing DNA vaccines for autoimmune arthritis.
Current therapies for rheumatoid arthritis (RA) and other autoimmune diseases non-specifically suppress immune function, and there is great need for fundamental approaches such as antigen-specific tolerizing therapy. In this paper we describe development of antigen-specific tolerizing DNA vaccines to treat collagen-induced arthritis (CIA) in mice, and use of protein microarrays to monitor respo...
متن کاملMucosal administration of IL-10 enhances oral tolerance in autoimmune encephalomyelitis and diabetes.
IL-10 is an immunoregulatory cytokine that can modulate immune processes, inhibiting the expression of inflammatory T(h)1 type responses as well as affecting antigen-presenting cell function. In addition, IL-10 has been shown to be active at mucosal surfaces. In the present study, we examined the role of IL-10 on orally and nasally induced tolerance. Treatment of (PL/J x SJL)F(1) mice with low-...
متن کاملRegulatory CD4+ T Cells Expressing Endogenous T Cell Receptor Chains Protect Myelin Basic Protein–specific Transgenic Mice from Spontaneous Autoimmune Encephalomyelitis
The development of T cell-mediated autoimmune diseases hinges on the balance between effector and regulatory mechanisms. Using two transgenic mouse lines expressing identical myelin basic protein (MBP)-specific T cell receptor (TCR) genes, we have previously shown that mice bearing exclusively MBP-specific T cells (designated T/R-) spontaneously develop experimental autoimmune encephalomyelitis...
متن کاملدرمان موشهای دیابتیک نوع 1 با آل- ترانس رتینوئیک اسید از طریق مهار سایتوکاینهای پیش التهابی
Background & Aims: Type 1 diabetes is an autoimmune condition associated with the T-cell–mediated destruction of Pancreatic β cells. Vitamin A (retinol) and its metabolites (such as all-trans retinoic acid (ATRA)) have a variety of biological activities including immunomodulatory action in a number of inflammatory and autoimmune conditions. The purpose of this study was to investigate the e...
